From the abstract: Here, we demonstrate efficient de novo synthesis of high G+C% DNA fragments using terminal deoxynucleotidyl transferase chemistry, the reconstruction of complete mycobacteriophage genomes using High-Complexity Golden Gate Assembly, and efficient rebooting via electroporation into Mycobacterium smegmatis.
